Yes you can cap the return on a mechanical fuel pump.

This will increase pressure and volume by about 10 to 15%. This usually doesn't cause any problems but on rare occasions can cause flooding when using a Q-jet carb.

Capping the return will increase the chance of vapor lock but not anymore so than running a mechanical pump that doesn't have a return.
