My current set up in my '64 GTO is the one I like the best.

Originally, I drove it with the stock 389, M20 and 3.23 open rear and 26.5 - 27" tires. Next I swapped in a 3.36 posi, which was eventually replaced with a 12 bolt 3.42 posi.

After building a 462 and driving it with the 3.42's, I decided that I wanted less rear gear with the bigger motor. Swapped the 3.42's for 3.08's.

This combo is the one I like the best as I do spend time on the Interstates going to shows (I go 4 hours one way every year). At some time in the future, I may consider a TKX with the wide ratio - 3.27 first gear and a .72 overdrive. This would help me out at the stop lights with the 3.27 first gear and on the Interstate with 2000 RPM at 70 MPH with the overdrive.
